Abstract
Monkeys with resting tremor produced by tegmental lesions, when given high doses of l-Dopa or a decarboxylase inhibitor plus l-Dopa, develop abnormal involuntary movements which closely resemble those seen in some Parkinson patients on l-Dopa therapy. These abnormal movements in the monkey may be subtle: slight turning of the head, lip smacking, tongue rolling, and chewing. Others may be extreme: marked grimacing associated with chorea-like movements of the head, upper and lower extremities; or with body swaying and peculiar posturing; such movements are dose dependent and disappear when the dosage is lowered. In the monkey, however, the dose of l-Dopa which relieves resting tremor, rarely produces abnormal movements. Such an analogue of abnormal movements produced by l-Dopa in monkeys with resting tremor, similar to that seen in some Parkinson patients on l-Dopa therapy affords a practical means of studying the mechanisms producing these abnormal movements, as well as a means for testing the effects of drugs on abnormal movements prior to clinical trial in man.
